Custom columns to auto-calculate and hold values of two other "number" columns, meaning that you have formula column adding/Subtracting... two other "number" columns... 
Case-in-point: The Favro "Sales CRM" template has a series of columns laid out: 
[Deal size] x [Close probability] = [Forecasted Value]... does not auto calculate!, meaning there is no formula calculating [Forecasted Value] 
So does that mean that someone has to manually calculate each record?

I want 3 separate number columns and Sum all of them in a 4:th column in sheet backlog.

Also being able to select which columns would be prefered almost like excel function =SUM(A1+A2+A3)

Add ability to calculate rank based on multiple parameters, which will help to prioritise. For example, on each story I should be able to specify: "Effort", "Risk", "Business Value" and have automatically calculated field "Rank" set by formula (Business Value + Risk)/ Effort

Different companies might have different formulas and it's components to rank the features

This would improve Favro to the next level. Almost like a small Excel inside Favro. Currently we are also running into this. Where we sometimes manually copy all the values over to Excel to do a simple sum or calculation there.
